光辐照度在反应器中的分布及光质对黄花蒿发根培养生产青蒿素影响的研究

中文摘要青蒿素是从传统治疟中药植物黄花蒿中提取的治疟有效成分,为世界卫生组织推荐使用的药品,在国际市场上供不应求。通过黄花蒿发根培育生产青蒿素可以克服野生资源不足,大田种植产量不稳定等缺陷。光因子对黄花蒿发根的生长和青蒿素合成均有很大的影响。本论文的研究了光辐照度在反应器中的变化规律,给出黄花蒿发根培养及青蒿素合成的最佳初始光辐照度为50-60μmol/m~2·s。反应器放大设计径向尺寸判据为植物组织器官与具有最佳初始光辐照度的光源的最远距离不得超过7cm。并在本实验的条件下,建立了青蒿发根生物量、青蒿素产量随光辐照度变化的数学模型。此外,对光辐照度随径向、轴向变化的规律也初步进行了数学描述。光质以红光最佳,在红光下青蒿发根生物量和青蒿素含量分别比白光下高17%和67%。
英文摘要The characteristics of light, including light irradiation and spectral quality, are very important to the growth of Artemisia annua L. hairy root and the production of artemisinin. It is necessary to study the distribution of light irradiation in a plant organ photobioreactor and the effects of light irradiation distribution as well as the spectral quality on the hairy root growth and the artemisinin production. Through studding the light irradiation distribution in the photobioreacter, the design criteria of a photobioreactor was obtained, i.e. the max distance from the hairy root to illuminant was 7cm. And it was concluded that the initial light irradiation ranged from 50 to 60μmol/m~2·s may be suitable to culture the hairy root in the photobioreactor. Also, a mathematical model describing the relations of hairy root biomass, artemisinin production and light irradiation distribution was build in the scope of this study. From the investigation of the effects of spectral quality, it was concluded that the red light with a 660nm peak value of wavelength was optimal to the growth of hairy root and the production of artemisinin comparing to the white, blue, yellow and green light.
