耐高渗酵母发酵法生产甘油的工艺优化--渗透压调节及细胞膜透性调节及流加发酵的工艺优化

中文摘要耐高渗酵母发酵法生产甘油是甘油生产的一条重要途径。目前发酵法生产甘油存在着发酵液中甘油/残糖比低而导致甘油提取困难、甘油产率低、生产成本高等问题。为了最大限度的提高菌体产甘油能力及后期耗糖速率,本论文做了三方面的工作:首先,本文通过在发酵过程的不同时间添加不同量的NaCl,研究了耐高渗酵母在不同渗透压条件下的生长代谢特点及不同时间加入不同量的NaCl对发酵过程的影响,发现适时添加适量NaCl造成的持续高渗环境有助于菌体产甘油,提高甘油产率及转化率;其次,研究了两种多烯类抗生素,两性霉素和制霉菌素对耐高渗酵母的生长代谢和对发酵过程各参数的影响,证明通过加入多烯类抗生素增加膜对甘油的透性从而提高甘油的产量是可行的,并将渗透压调节和细胞膜透性调节结合起来,更大的提高了甘油浓度;最后,将流加发酵中的葡萄糖浓度和渗透压水平联系起来,研究了初始糖浓度、投入糖总量、最适维持糖浓度、氮源和磷源对发酵过程的影响,得出了在现有条件下的最佳流加方案。
英文摘要To produce glycerol by fermentation with osmotolerant yeast is one of the most important methods in glycerol production. At present, there exists an intractable problem in this process: the low glycerol/glucose ratio, which can lead to the difficult extration of glycerol from broth, low yield and high cost. Three parts of work have been finished in this thesis in order to obviously increase the glycerol concentration in broth: Firstly, the physiological and metabolic behavior of osmotolerant yeast under media with different osmotic pressure and the influence of NaCl on fermentation process have been investigated by adding different amounts of NaCl at different fermentative stages. It's found that the continuing hyperosmotic environment by adding appropriate amount of NaCl at proper time can help the yeast to synthesize more glycerol, which can increase the rate of production and conversion yield. Secondly, the effect of two kinds of polyene macrolides, amphotericin B(AmB) and nystatin, on the growth and metabolism of osmotolerant yeast and on the fermentation process have been studied. It's proved that raising the glycerol output by increasing the membrane permeability with polyene macrolide is wokable. More glycerol can be produced by adding NaCl and polyene macrolides in the same fermentative media than adding one of them. Finally, by relating the glucose concentration with the osmotic pressure in broth, it was investigated that the influences on the fermentation process of initial glucose concentration, the total glucose quantity, the maintaining glucose concentration, the nitrogen and the phosphor and an optimal fed-batch scheme was determined.
