中文摘要本文首次研究了我国传统名贵中药材水母雪莲细胞悬浮培养生产黄酮类活性化合物的工艺过程。采用目视法和诱变处理,筛选到高产稳产愈伤组织细胞系。首次进行了水母雪莲细胞悬浮培养,建立了悬浮培养细胞系,确定了细胞悬浮培养的最适培养条件和最佳培养基组成,并采用真菌诱导子对细胞黄酮合成进行调控,使得水母雪莲悬浮培养细胞的黄酮含量超过了野生植株。采用2升搅拌式生物反应器进行了细胞大规模培养,初步建立了水母雪莲细胞反应器悬浮培养工艺。系统研究了水母雪莲细胞悬浮培养的动力学,建立了可以描述植物细胞培养中细胞生长的全部五个阶段的唯象动力学模型。
英文摘要Saussurea medusa Maxim, family Compositae, is well known in Chinese medicine, and now, it is almost extinct because of uncontrolled collection of the wild plants and the difficulty of cultivating it. So, there has been considerable interest in investigating the potential of plant cell cultures as an alternative for the industrial production of secondary metabolites. After having treated with 4000 rad of Co-60 γ-ray, a callus line which produced more flavonoids was selected. Flavonoid production was dramatically affected by light irradiation. Blue light, white light and far red light stimulated the PAL activity and flavonoid formation. The flavonoid accumulation of callus cultures increased with the increasing intensity of illumination under white light. The cell suspension cultures of S. medusa was firstly established from callus cultures. The flavonoid content varied greatly with the change of cell aggregate size, and cell aggregates ranged from 1.0 mm to 5.0 mm had the highest flavonoid content. Effects of some factors on the degree of cell aggregation were studied. The optimum inoculum concentration, pH value of the medium and medium composition for cell aggregate suspension cultures of S. medusa were were all determined. Precursors and fungal elicitors were fed to improve flavonoid accumulation in cell aggregate suspension cultures of S. medusa. The result showed that the elicitor extracted from Aspergillus niger was most effective of the elicitors used. Cell aggregate suspension cultures of S. medusa were successfully carried out in a 2-L stirred bioreactor. These results might be useful for the scale-up of flavonoid production. The kinetics of cell aggregate suspension cultures of S. medusa was studied comprehensively. The synthesis of flavonoids was growth associated. Yield coefficients of sucrose and nitrogen were also calculated from experiment data. The phenomenological kinetic model of cell growth was established based on the ecological theory. The regression results fit well with the experiment data. This phenomenological model could provide a good description of cell growth in cell aggregate suspension cultures of S. medusa.
