辣根过氧化物酶的次生代谢调控和反应器技术研究

关键词辣根过氧化物酶(HPR)是广泛应用于临床诊断和微量分析的免疫测定试剂用酶。本文研究了通过组织细胞培养方法生产辣根过氧化物酶次生代谢物调控和生物反应器技术。为大规模植物组织培养生产辣根过氧化物酶提取供工程基础依据。论文研究了:(1)植物生长因子对辣根菜愈伤组织诱导、组织分化和高产系筛选区的影响 同时利用农杆菌基因转导方式,将Ti、Ri质粒基因转入植物细胞中,诱导冠瘿瘤愈伤组织和发根产生,研究化学诱导信号AS和遗传转化过程操作条件对质粒转达化率先的影响。(2)根团的培养工艺及次生代谢产物调控--接种细胞形态、外源蛋白质添加、pH值调控、低温间歇刺激、光敏因素、培养方式等条件调控次生代谢产物合成。(3)分化根培养过程中的营养物质传递探索,研究了高密度根团培养内部的物质传递传质特点以及对生长和次生代谢产物合成影响。(4)植物细胞培养反应器技术,比较了浸没、非浸没式生物反应器在植物组织培中的应用。

英文摘要Peroxidase is widely used as an important reagent for clinical diagnosis and microanalytical immunoassay. This research work focus on the metabolism regulation of the peroxidase production by tissue culture of orseradish and bioreactor technology. The research mainly contains four parts:(1)Plant tissue culture technique, selection of high-yield plant cell by plant regulators and Ri,Ti plasmid transformation of Agrobacterium (2)metabolism regulation and culture techniques of horseradish differential roots. Culture techniques such as inoculation morphology, supplement of casein hydrolysate acid(CHA), pH and control, temperature control, concentrations of Fe~(2_) and light irradiation conditions, affect the growth and metabolism of differential root. 93)Studies on mass transfer in high density culture of horseradish differential root mats. Oxygen transfer had an important effect on differential root cultures. (4) Application of bioreactor technology in horseradish cell and tissue culture. Comparing to the different type bioreactors: completely submerged bioreactor, incompletely submerged bioreactor and unsubmerged bioreactor, incompletely submerged bioreactor and unsubmerged bioreactor have some advantages, such as lightening vitrification, improving mass transfer and supplying better grown environment for plant cells.
