口蘑属的分子系统发育及中国该属的物种多样性研究

英文摘要Tricholoma (Tricholomataceae, Agaricales), a fungal genus widely distributed in different continents of the world, is rich in species diversity and is ecologically and economically very important. Currently available studies, which were conducted based mainly on morphological observations and single gene phylogenetic analyses, indicated that Tricholoma can be divided into 11 sections. However, these researches failed to clearly elucidate the phylogenetic relationships among these sections. Meanwhile, limited studies on the Chinese species diversity of Tricholoma are available to date. In this study, we collected samples from China and other countries, and carried out molecular phylogenetic analyses on these samples by using a multiple-gene strategy. A total of 60 DNA fragments were selected from the 96 single-copy genes available for the Agaricomycetes, which were later amplified and sequenced using the Next Generation Sequencing (NGS) procedure. With these data, a clear phylogenetic framework for Tricholoma was constructed, the phylogenetic relationship between the main branches of Tricholoma was analyzed for the first time. In addition, a preliminary survey on the species diversity of this genus in China was conducted and two new species were formally described. The main results are as following: 1. Phylogenetic framework of Tricholoma In total, 255 specimens from both China and abroad were used to amplify and sequence the 60 single-copy genes selected. After quality control, 5822 sequences of 45 genes were generated from 149 samples. Using both Mostly Likelihood (ML) and Bayesian Inference (BI) strategies, a clear phylogenetic framework of Tricholoma was constructed by using the concatenated dataset of these sequences. The phylogenetic trees revealed 11 sections, with sect. Pardinicutis as the basal group and the clade formed by sect. Lasciva and sect. Sericella as the sub-basal group. The relationships among sect. Caligata, sect. Muscaria, sect. Genuina, sect. Rigida and sect. Tricholoma were also elucidated. By using both morphological and molecular evidence, the members with sect. Megatricholoma were placed into sect. Genuina. In addition, a putative new section, which contains only samples collected from China, was uncovered. By comparing with previous studies, we discussed the key characters of each section. 2. Species diversity of Tricholoma in China We found that Tricholoma species can be found from northeastern, southwestern, northwestern and central China and the southwestern part harbors the richest species diversity in China. Specimens of over 70 species were gained and examined, of which over 40 species are known species and over 30 are putative new species. Two new species were described based on both morphological and molecular evidence. Other putative new species still need to be confirmed and described with further in-depth studies.
