Hypersampsone P抑制脂肪细胞生成的作用及其机制初探

英文摘要The obesity is increasing sharply in recent years. It became a hot problem of global concern for inducing diabetes, cancer, hypertension and other diseases. At present, the treatment of obesity mainly consists of lifestyle adjustment, surgical treatment and drug treatment, yet they all have deficiency. So finding safe and effective natural products as a lead compound for treating obesity is a long way off. But in the body obesity production mainly relates with the white fat deposition. White fat deposition mainly has two forms: The increase of adipocyte number and the increase of single adipocyte volume. The increase in the number of fat cells is also known as hyperplastic obesity, refers to the process of differentiation of preadipocytes into fat cells. At present physical exercise has no therapeutic effect on this kind of obesity. Adamantane polycyclic polyprenylated acylphloroglucinols (PPAP), a special class of hybrid natural products with caged architecture widely exists in plants, especially Hypericum. Hypersampsone P is a PPAP compound extracted from Hypericum subsessile. Currently, few studies have been conducted on the pharmacological activity of amantadine PPAP, and its pharmacological activity of inhibiting adipocyte differentiation has not been reported. Here we applied the 3T3-L1 adipocyte model to screen ten PPAP for their activities in adipocyte differentiation. The result shows that hypersampsone P (HSP) inhibited adipocyte differentiation significantly with a EC50 10.93 μM; and the effect is most likely related to the configuration of C-5 and the substituent type of C-28, because the other two compounds, similar to hypersampsone P, hypersampsone H and hypersampsone M did not affect the process of adipogenesis. The expressions of key regulators for adipogenesis, PPARγ, FABP4 and SREBP1c were down-regulated by HSP. However, the AKT and AMPK signal pathway was not affected by HSP at the terminal of differentiation, whereas the phosphorylation of LKB1S428 and AMPKT172 was enhanced, the gene expression of downstream SREBP1c, ACCα, SCD1 and FAS were downregulated in the early stage of differentiation. In conclusion, hypersampsone P is capable of enhancing AMPK and inhibiting of adipocytes differentiation as the consequence, indicating its mother-core structure could be a candidate for development of drugs for treatment of obesity.
