利用 Q-TOF LC/MS 对五味子科植物次生代谢产物库的初步构建

英文摘要The research on the chemical constituents of Schisandraceae species has been lasted for over 30 years in our research group, and a compound library containing compounds obtained from these species has been built. In order to extend the capability of this compound library to metabolite profiling of mixtures, isolation of specific compound, dereplication, metabolomics, etc., to build a sub-library containing the spectroscopic data of compounds will provide important research foundation and support. The current research preliminarily explored the creation of a mass spectra library for compounds obtained from Schisandraceae species, and examined the value of this library in metabolite profiling of mixtures. Besides, in order to enrich and diversify the compounds and mixtures necessary for the building of the mass spectra library, as well as to carry out preliminary discussion on specific cases, extensive investigation on the chemical constituents of Schisandra rubriflora, a plant species with good medicinal value, was also undertaken. Notablely, the plant material under research was grown in special inhabitant. In-depth research on Schisandra rubriflora (Schisandraceae) collected from Qiaoshan village, Yunnan Province, Hubei province, Dali Huatian Dam have been previously carried out and proved this species to be rich in bioactive constituents, including schinortritepenoids (SNTs), lignans, etc. It is well known that the inhabitants of plants have great influence on their secondary metabolism, thus, S. rubriflora collected from Medog County, Tibet was subjected to systematic phytochemical research. As a result, 61 compounds were characterized and 9 of them were previously unreported. These compounds involve SNTs, lignans, triterpenoids, and sesquiterpenes. Bioactivity evaluation of several new compounds were undertaken, which revealed that rubrischidilactone A (1) (a SNT) and rubrischilignan B (a dibenzocyclooctadiene lignan) can promote the cell differentiation of PC12 cells. The ultra-high performance liquid chromatography-quadrupole time-of-flight mass spectrometry (UHPLC/Q-TOF-MS) is a rapid and efficient technique of liquid chromatography-tandem mass spectrometry, and it can accomplish the challenging qualitative and quantitative analysis with high resolution and sensitivity. In order to preliminarily study the creation of a mass spectra library for compounds obtained from Schisandraceae species, the current research collected the accurate mass, retention time and MS/MS data of 9 compounds obtained from Schisandra rubriflora, and PCDL Manager was then used to process these data to create the library. To preliminarily examine the application of this library in metabolite profiling of mixtures, several extracts of Schisandra rubriflora were also subjected to Q-TOF analysis.
