芳樟精油改善抑郁小鼠行为和海马可塑性的分子机制

中文摘要Objective: Depression is a global mental health problem, characterized by recurrent depressive episodes, depression, lack of interest and pleasure, accompanied by anxiety, sleep and appetite disorders, which not only affects patients' social function and quality of life, but also may lead to a variety of diseases. The increasing incidence of depression leads to the rising social and economic burden in the world, which is an urgent problem to be solved in modern society. The purpose of this study is to discuss the antidepressant effect of Cinnamomum camphora (Linn.)Presl essential oil in aromatherapy as a supplementary therapy for depression, its influence on inflammatory factors TNF-α, IL-6 and IL-1β, and its regulatory mechanism on Nrf2/HO-1 pathway. Methods: The depression model of mice was established by intraperitoneal injection of lipopolysaccharide(LPS), and behavioral tests were carried out by open field test, elevated cross maze test and forced swimming test. Western blot and qRT-PCR were used to detect the expression level of Nrf2/HO-1 pathway protein in hippocampus of mice, enzyme-linked immunosorbent assay (ELISA) was used to detect the changes of inflammatory factors TNF-α, IL-6 and IL-1β in serum, and Tunel staining was used to detect the changes of apoptosis in brain tissue of mice. Results: The results showed that compared with the blank control group, the movement distance, the stay time and the times of entering the central area of the model group were significantly reduced, the percentage of time (OT%) and the percentage of times (OE%) of entering the open arm in the elevated cross maze experiment were significantly reduced, and the immobility time of the forced swimming experiment mice was significantly prolonged. Compared with the model group, after the treatment of Cinnamomum camphora(Linn.)Presl essential oil, the moving distance, the staying time and the times of entering the open field center increased significantly, the OT% and OE% of mice entering the open arm in the elevated cross maze increased significantly, and the immobility time of mice in the forced swimming experiment decreased significantly. In addition, the expression levels of Nrf2 and HO-1 protein in the model group decreased significantly, while TNF-α, IL-6 and IL-1β inflammatory factors increased significantly. After the treatment of Cinnamomum camphora(Linn.)Presl essential oil, the expression levels of Nrf2 and HO-1 protein in depressed mice were significantly increased, and the inflammatory factors TNF-α, IL-6 and IL-1β were significantly decreased. The results of Tunel staining showed that the apoptosis rate of brain tissue cells in mice decreased significantly after the treatment of Cinnamomum camphora(Linn.)Presl essential oil. Conclusion: Cinnamomum camphora(Linn.)Presl essential oil can effectively improve the depression-like behavior of mice, and its mechanism may be related to its participation in regulating Nrf2/HO-1 signaling pathway, up-regulating the expression levels of Nrf2, HO-1 protein and mRNA, and preventing the inflammatory injury of hippocampus. This discovery provides empirical support for the treatment and application of Cinnamomum camphora(Linn.)Presl essential oil and aromatherapy in depression
英文摘要目的:抑郁症是一种全球性的心理健康问题,表现为反复的抑郁发作、情绪低落、兴趣缺乏和快感缺失等,同时伴有焦虑、睡眠和食欲紊乱等症状,不仅影响着患者的社会功能和生活质量,还可能导致多种疾病的发生。日益增长的抑郁症发生率导致全球的社会负担和经济负担不断攀升,是现代社会亟待解决的问题。芳香疗法是以芳香精油为主要治疗药物的一种补充与替代疗法,多种精油经研究证实对抑郁症有显著改善作用,重要的精油成分之一芳樟醇有很好的抗炎抗抑郁的功效。本研究旨在研究中国特色树种芳樟的主要经济产物芳樟精油的抗抑郁作用及其对小鼠海马炎症因子 TNF-α、IL-6 和 IL-1β的影响以及其对 Nrf2/HO-1 通路的调控机制。 方法:采用腹腔注射脂多糖(lipopolysaccharide, LPS)建立小鼠抑郁模型,利用旷场实验、高架十字迷宫实验和强迫游泳实验进行行为学实验;通过 Western blot 和 qRT-PCR 方法检测小鼠海马组织中 Nrf2/HO-1 通路蛋白表达水平变化, 用酶联免疫吸附检测(ELISA)法检测海马组织中炎症因子 TNF-α、IL-6 和 IL-1β 的含量变化,并通过 Tunel 染色检测小鼠海马脑组织细胞凋亡情况的变化。 结果:结果表明,与空白对照组相比,各组小鼠旷场总运动距离没有显著 性差异,说明各组小鼠没有因为药物的使用影响自发性运动;模型组小鼠旷场 中心区运动距离、旷场中心的停留时间和中心区域的进入次数显著减少,高架 十字迷宫实验中小鼠进入开臂的时间百分比(OT%)和进入开臂的次数百分比 (OE%)显著减少,强迫游泳实验小鼠不动时间显著延长,说明 LPS 引发了小鼠的抑郁样状态;而相较模型组,给予芳樟精油治疗后,小鼠在旷场实验中心区域运动距离显著增加,在旷场中心区域的停留时间显著增加,旷场中心区域的进入次数显著增加,高架十字迷宫小鼠进入开臂的 OT%和 OE%显著增加,强迫游泳实验中小鼠不动时间显著减少;此外,生物学检测结果显示,与空白对照组相比,模型组小鼠的 Nrf2、HO-1 蛋白和 mRNA 表达水平显著下降,TNF-α、IL-6 和 IL-1β 炎症因子显著升高,小鼠海马细胞凋亡率显著升高;给予芳樟精油治疗后能显著提高抑郁模型小鼠 Nrf2、HO-1 蛋白和 mRNA 表达水平,并显著降低炎性因子 TNF-α、IL-6 和 IL-1β 水平,小鼠脑组织细胞凋亡率也显著下降。 结论:综上所述,芳樟精油能有效改善小鼠的抑郁样行为,其机制可能与其参与调控 Nrf2/HO-1 信号通路,上调 Nrf2、HO-1 蛋白和 mRNA 表达水平并阻止海马炎症损伤有关,这一发现为芳樟精油及芳香疗法在抑郁症中的治疗及应用提供了实证支持。
