Differential recognition and targeting isolation of HPLC co-efflux isomers using countercurrent chromatography based on LC-MS feature analysis

英文摘要 | Identifying and separating of high-performance liquid chromatography (HPLC) co-efflux isomers using conventional column chromatography with a solid phase as the carrier to obtain pure compounds are quite challenging because the isomers have similar polarities and their contents vary. Countercurrent chromatography (CCC) and feature analysis based on liquid chromatography-mass spectrometry were employed to efficiently enrich and separate HPLC co-efflux isomers. Based on the core fragments of co-efflux isomers produced during in- source collision-induced dissociation (CID) process, the discrepant features of the core fragments were recognized and quantified. The precise partition coefficients (K) K ) of co-efflux isomers were obtained via nontargeted and targeted LC-MS analysis. Thus, HPLC co-efflux isomers present in multicomponent systems could be efficiently analyzed, enriched, and isolated. A solvent system comprising n-hexane:ethyl acetate:methanol:water (1:5:1:5; v:v:v:v) was selected for use in CCC. Four flavonoids containing three isomers were isolated from Rheum tanguticum leaves. The feature recognition and targeted isolation of HPLC co-efflux isomers could therefore be achieved using the proposed novel method. |
