Complete mitochondrial genomes and population genetic analysis of Brachidontes variabilis (Krauss1848) in China

英文摘要Brachidontes variabilis (Krauss, 1848) is a common small mussel widely distributed in the intertidal zone of southern China. Firstly, we characterized four complete mitochondrial genomes of B. variabilis from four populations (Wanning, Yangjiang, Zhangzhou, and Shanwei) in Chinese waters. This study was the first to sequence and discribe the mitochondrial genome of B. variabilis, which comprised of 2 ribosomal RNAs (rRNAs), 22 transfer RNAs (tRNAs) and 12 protein-coding genes (PCGs, lacking atp8). And then three PCGs fragments, including cytochrome oxidase subunit I (cox1, 558 bp), NADH dehydrogenase subunit 5 (nad5, 754 bp), and NADH dehydrogenase subunit 4 (nad4, 879 bp), were utilized to investigate the population genetics and demography. The cox1, nad5 and nad4 fragments revealed 55, 81, and 103 haplotypes, respectively. Results indicated high genetic diversity within populations (Hd > 0.5, Pi > 0.005) and little genetic differentiation among populations (F-ST<0.05), suggesting frequent gene exchanges. Further, the neutrality test and Extended Bayesian skyline plots (EBSPs) suggested that the four populations underwent population expansion at 0.9-1.1 Ma during the Pleistocene. In summary, this study on the mitochondrial genomes and population genetics of B. variabilis provides insights into the evolutionary history and demographic dynamics of this species in southern China.
