Arabinogalactan-like polysaccharide isolated from the flowers of Dendrobium officinale regulates the gut microbes and promotes vitamin K1, vitamin A, and vitamin B6 products

| 英文摘要 | Arabinogalactan (AG) is one ubiquitous feature of plant polysaccharides that may regulate Human Gut Micro-biota (HGM). However, whether AG induces vitamin generation linked to HGM modified by this polysaccharide is still vague. We hypothesize that the gut microbiota possesses the ability to degrade AG, while AG may reshape HGM to produce vitamins. Here, we showed that an AG-like polysaccharide TF112 (Mw 54.7 kDa) was purified from the flowers of Dendrobium officinale. It comprised rhamnose, galacturonic acid, galactose, and arabinose in a molar ratio of 1.00: 1.01: 4.00: 2.55. The backbone was an alternating 1, 2, 4-linked Rha and 1, 4-linked GalA, where the O-4 position of 1, 2, 4-Rha was substituted by three types branches: one as-*1)-Gal-(4-*1)-Gal, another as-*1)-Ara, and the remaining one as arabinogalactan polymers, respectively. We further showed that TF112 can be degraded into monosaccharides and oligosaccharides by Bacteroides spp. Further, we provided evidence that TF112 changed the composition of the gut microbiota of mice and increased the content of vitamin K1, vitamin A, and vitamin B6 in the feces of mice. Overall, our research provides new insights into understanding the crosstalk between AG and HGM linked to vitamin generation. |
