Sea buckthorn triterpenic acids alleviates metabolic dysfunction-associated steatotic liver disease via modulation of liver biochemistry, lipid metabolism and AMPK/Nrf2/NF-κB signaling pathway

英文摘要Sea buckthorn triterpenic acids (STE) have been shown to provide health-promoting benefits in alleviating metabolic dysfunction-associated steatotic liver disease (MASLD). However, the specific impact on liver and underlying regulation mechanisms has not yet been fully elucidated. The MASLD animal model was established through administration of high-fat diet to C57BL/6J mice for 12 weeks. The mice were given STE at the doses of 100 and 300 mg/kg for 12 weeks. The experimental findings indicated that STE reduced the body and liver weights of MASLD mice and alleviated the biochemistry indicators of liver function, lipid accumulation, oxidative stress and inflammation. Histopathological results showed that STE mitigated lipid accumulation, hepatic steatosis and mitochondrial injury. Liver lipidomics analysis revealed that STE could significantly alter the changes in lipid metabolism spectrum in MASLD mice, including modulation of triglyceride, diacylglycerol, phosphatidylethanolamine and ceramides. Furthermore, STE regulated the protein expression of lipid metabolism, oxidative stress and inflammatory response through the AMPK/Nrf2/NF-kappa B signal pathway. In conclusion, this study demonstrates STE could serve as a potential functional food ingredient and offer new idea for the prevention and treatment of MASLD.
