Distinct genetic groups with restricted gene flow in wild crop relatives Saccharina cichorioides from the northwestern Pacific

| 英文摘要 | Saccharina cichorioides is an important wild germplasm resource for the breeding of cultivated kelp. However, due to the limited understanding of its population structure and genetic diversity, the direct application in crossbreeding can lead to randomness and unpredictability, ultimately increasing costs and duration of the breeding process. To address these challenges, a population genetics analysis was performed on 13 populations of S. cichorioides using the mitochondrial marker COI and 9 microsatellite markers. Our study suggested that S. cichorioides could be classified into two distinct groups: the group OKS (populations existed along the east coast of Sakhalin Island), and the group JPS (populations located along the shoreline of the Sea of Japan). The JPS group exhibited a higher level of genetic diversity, as evidenced by significantly higher values in parameters such as haplotype diversity (Hd = 0.641), observed heterozygosity (HO = 0.357), and expected heterozygosity (HE = 0.517). However, Approximate Bayesian Computation (ABC) simulations indicated that the OKS group might represent ancient populations. Gene flow analysis revealed that there was limited genetic exchange between the two groups, which might be attributed to the geographical isolation in populations of S. cichorioides. Collectively, our study of the genetic diversity and population structure of S. cichorioides enriches our understanding to its population patterns, and could provide guidance for the collection of wild germplasm and resources protection. |
