SsGSDMEa-mediated pyroptosis is involved in Photobacterium damselae subsp. piscicida infection in the black rockfish Sebastes schlegelii

英文摘要Pyroptosis is a necrotic cell death mediated by gasdermin, which is activated by caspase (CASP) cleavage. CASP also mediates apoptosis, a non-necrotic cell death. Photobacterium damselae subsp. piscicida (PDP) is a bacterial pathogen that causes necrotic cell death in fish, including black rockfish Sebastes schlegelii, but the underlying mechanism, especially that associated with pyroptosis, remains unclear. Here, using S. schlegelii as a representative species, we examined the involvement of gasdermin-mediated pyroptosis in PDP infection in fish. We identified two gasdermin E (GSDME) orthologs, SsGSDMEa and SsGSDMEb, from S. schlegelii and found that only SsGSDMEa could trigger pyroptosis. We also identified three S. schlegelii CASPs (SsCASP3, 9, and 10), all of which induced apoptosis. SsGSDMEa was cleaved by SsCASP3 at 261NVVD264, producing an N-terminal fragment (NT) named NT264. SsGSDMEa was also cleaved by SsCASP 9 and 10 in the same manner at 203IESD206 and 246IEAD249, producing NT206 and NT249, respectively. NT264 and NT249, but not NT206, triggered pyroptosis. PDP infection upregulated SsGSDMEa and SsCASP3/9/10 expressions in fish tissues and activated SsGSDMEa and SsCASP3/9/ 10 in fish cells, leading to pyroptosis. Taken together, the findings deepened our understanding of the CASPmediated activation mechanism of black rockfish GSDME and its involvement in pathogen infection.
