Applying natural product repurposing strategy to identify baicalein as novel caseinolytic protease P inhibitor and its application in the treatment of rice bacterial diseases

| 英文摘要 | Plant diseases caused by bacteria affect the yield of crop, greatly reduce the quality of food, and thus posing a great threat to food safety. To fill the gap that no report about ClpP inhibitor is applied in agri-food production field, engineering natural-product repurposing strategy, 55 of natural products were screened using the combination of ClpP inhibitors of Xanthomonas oryzae pv. oryzae (Xoo) screening assay and anti-Xoo activity experiment. Outcomes showed that naturally occurring baicalein possessed favorable peptidase inhibitory activity against XooClpP (IC50 = 130.32 mu M) and in vitro anti-Xoo activity (EC50 = 7.11 mg L-1). Transcriptome analysis indicated that baicalein directly interacted with XooClpP, reduced the stability of XooClpP, further inhibited intracellular processes including oxidative phosphorylation and tricarboxylic acid cycle, and thus kill bacteria. Overall, this study highlights interesting insights into baicalein as a new inhibitor of XooClpP, and shows its unprecedented prospect as new bactericide candidate for ensuring the crop health and food safety. |
