NIR-II organic fluorophores integrated with blood-brain barrier-penetrating strategies for brain disease theranostics

| 英文摘要 | The precise diagnosis and effective treatment of brain diseases have long posed significant challenges in the field of neuroscience. In recent years, near-infrared II (NIR-II, 1000-1700 nm) fluorescence imaging has emerged as a revolutionary tool for high-resolution in vivo brain imaging, owing to its advantages such as deep tissue penetration and minimal autofluorescence. This review systematically summarizes recent advances in NIR-II organic fluorophores applied in the diagnosis and treatment of brain disorders, with particular emphasis on molecular design strategies and optimization of optical properties. Moreover, the key to achieving effective diagnosis and therapy lies in the fluorophores' ability to efficiently cross the blood-brain barrier (BBB). Therefore, this work explores various active targeting and stimulus-responsive delivery strategies developed to overcome the BBB. Furthermore, we highlight the applications of these functionalized NIR-II organic probes in brain disease imaging and therapeutic visualization. Finally, we discuss current challenges and future perspectives in this field, aiming to provide insights for advancing the application of NIR-II fluorophores in precision medicine for brain diseases. |
