Advances in fungal vaccine development: Progress in the face of emerging challenges

| 英文摘要 | Historically overshadowed by bacterial and viral infections, fungal diseases are resurging globally, fueled by rising antifungal resistance and the increasing vulnerability of immunocompromised populations in healthcare settings. The widespread misuse and overuse of antifungals have accelerated the emergence of resistant fungal pathogens, complicating clinical management. Despite significant progress in vaccine development, with candidates such as NDV-3A and PEV7 advancing to late-stage trials, none have received FDA approval. Consequently, novel, more effective, and tolerable antifungal therapies are urgently needed. This review consolidates data on the progress made in fungal vaccine development, emphasizing key obstacles to clinical realization. It further proposes innovative strategies, such as bioinformatics-driven antigen discovery, nanotechnology-based delivery systems, reverse vaccinology, and the exploration of probioticvaccine synergies, which hold exceptional promise in overcoming these challenges and advancing the fight against fungal infections. |
