中文摘要反胶团萃取分离蛋白质是一种新型、非常有发展前途的生物物质分离方法。本文从反胶团萃取分离蛋白质的过程机理到反胶团在实际应用中的可行性等多个方面,较全面系统地研究了反胶团法萃取分离蛋白质。阳离子表面活性剂Aliquat 336是一种含混合碳链长度的季铵盐,在加入多种直链醇作助溶剂时,它可溶于多种非极性的溶剂中而形成反胶团。本文进行了用Aliquat 336与异辛烷及正丁醇构成的反胶团萃取α-淀粉酶发酵液的实验,发现如果能将发酵液适当地加以稀释,就可利用反胶团溶液直接从发酵液中分离提纯α-淀粉酶,而且还可以大幅度地去除发酵液中存在的中性蛋白酶。
英文摘要The reversed micellar extraction is a new and rapidly developing method of separating biomaterials, expecially on protein. In this paper, a systematic study on this subject from the aspect of the mechanism of reversed micelles extraction of protein to the aspect of the devlopment of this technique to the industrial practice was carried out. Aliquat 336 is a quaternary ammonium salt cationic surfactant. With alcohol as co-solvent, it can form reversed micelles in many apolar solvents. The reversed micelles of Aliquat 336/isooctane/n-butanol were also used to extract the fermentation broth of α-amylase. The results demonstrate that the reversed micelles can be used to purify the α-amylase from the fermentation broth if the broth is diluted first to decrease its ionic strength. The reversed micellar solution can recover a very large amount of enzyme activity and remove quite a large amount of the harmful neutral protease in the broth at the same time.
