Identification of Two Novel O2-Conotoxins from Conus generalis

| 英文摘要 | Conus generalis is a type of worm-hunting snail commonly found in subtidal habitats. Fractionation of its venom extract revealed two main peaks overwhelming in the intensity. In this work, by integrating mass spectrometry, N-terminal amino acid sequencing, and molecular cloning, the first dominant fraction was identified as two novel conotoxins, designated ge6b and ge6c, which originate from the same precursor but differ by one gamma-carboxylation modification. These two conotoxins are the first natural peptides identified from C. generalis, and they were classified as members of the O2-superfamily based on the signal sequence. Intracranial injection of the reconstituted fraction into mice induced depressive symptoms, from which the mice recovered after 10-15 min. However, no obvious current change was detected in response to ge6b/c on voltage-gated Na+, K+, or Ca2+ channels, or on nicotinic acetylcholine receptors (nAChR). These results suggest that these two conotoxins may exert their effects through a distinct membrane of channel or receptor. |
