片仔癀对小鼠急性酒精性肝损伤的干预研究

中文摘要片仔癀是我国知名的传统名贵中成药,至今已有450余年的应用历史,临床上广泛用于治疗各种急性、慢性、病毒性肝炎。近年来我国由于过度饮酒引起的酒精性肝病发生率呈明显上升趋势,严重威胁国民健康。本课题中我们采用小鼠急性酒精肝模型探索片仔癀对急性酒精性肝损伤的干预效果。通过对小鼠进行为期两周的含酒精的液体饮食诱导(含5%酒精的液体粮食饲喂3天后,继续饲喂含8%酒精的液体粮食11天),饥饿过夜后于处死前6小时进行5g/kg酒精灌胃刺激,造成小鼠急性酒精性肝损伤模型。酒精饮食诱导期间实验组小鼠每12小时给予高剂量(0.5mg/g)、低剂量(0.1mg/g)的片仔癀水溶液灌胃一次。结果显示,低剂量片仔癀治疗能显著性抑制酒精所致的血清ALT、AST水平升高,及肝脏甘油三酯含量增加;减少肝组织切片中的空泡数目,降低肝组织中的脂滴积累;并显著抑制糖脂代谢中关键基因PPARγ及促炎因子IL-β 和MCP-1的表达水平。与此相反,高剂量片仔癀治疗显著性加重了酒精所致的肝脏炎症反应及脂肪变性,但对酒精所致PPARγ、IL-1β 和MCP-1的表达水平的影响与模型组相比无显著性差异。这一结果说明,在一定剂量下片仔癀对酒精性肝损伤具有保护作用,但过量的片仔癀干预却会加重酒精所致的肝脏炎症及脂肪变性。故若使用片仔癀用于酒精性肝病预防与治疗需在经验丰富的临床医生的指导和监控下进行。关键词:急性酒精性肝损伤、片仔癀

英文摘要Abstract:Pien Tze Huang (PZH), a formula of traditional Chinese medicine, has been extensively used for treating acute, chronic or viral hepatitis. We took this study to investigate the potential activity of it in protecting against acute alcohol-induced liver injury in mice. Alcoholic liver injury was achieved by feeding mice with a liquid diet containing 5% ethanol for 3 days and then 8% ethanol for 11 days, followed by a single dose of ethanol gavage(5g/kg). PZH was administered every 12h at a low dose (0.1 mg/g body weight/dose) and a high dose (0.5 mg/g body weight/dose). The alcohol-induced increase of the serum ALT and AST levels were significantly abrogated by low dose PZH but aggravated by high dose PZH. Besides, histological staining revealed that low dose PZH reduced alcohol-induced lipid accumulation in the liver, but high dose PZH promoted liver steatosis. Furthermore, low dose PZH ameliorated alcohol-induced elevation of adipogenesis gene PPARγ and proinflammatory factors IL-1β and MCP-1.Collectively, these data indicate that alcohol-induced liver injury and steatosis were significantly ameliorated by low dose PZH but aggravated by high dose PZH.
