Several problems in analysis of flavonoids from Ginkgo biloba by high performance liquid chromatography

| 英文摘要 | the operating conditions in several key links for analysis of contents of flavonoids from the leaves of ginkgo biloba by high performance liquid chromatography( hplc) have been investigated, they included the composition of hydrolysis solution, the temperature and the time of the hydrolysis. the possibility of using only one standard of quercetin to evaluate the contents of three flavonoids, i.e. quercetin, kaempferol and isorhamnetin at a wavelength of 370nm was demonstrated. the selected hydrolysis conditions were using of a 5.5%( w/v) hcl in methanol as hydrolysis solution at 85 similar to 90 degrees c oil bath where temperature hold slight boiling and to reflux for 1.3 hrs. hplc conditions were bio-sil ods,5 mu m, column, 150 x 4.0 mm i.d., meoh/0.4%( v/v) phosphoric acid aquious solution(57: 43) as the mobile phase and detection at 370 nm. the satisfactory recovery and analytical accuracy can be obtained under these operating conditions. |
