Enantiomeric separation of amino acid derivatives by ion-pair chromatography

| 英文摘要 | the enantiomeric separation of n-3,5-dinitrtobenzoyl (dnb) of amino acid derivatives by ion-pair chromatography with a chiral stationary phase (csp), derived from (s)-n-(2-naphthyl)-alanine has been examined. it was found that the increase in the concentration of a chiral ion-pair reagent caused significant increase in the capacity factors of both enantiomers of dnb-phenylglycine and dnb-leucine until a limiting value is reached, while the enantioselectivities of both analytes remained unchanged. a mobile phase of methanol-aqueous phosphate buffer was used. it was found that the concentration of methanol in the mobile phase dramatically affected the retention of the enantiomers. a minimum in enantioselectivity occurred at about 60% methanol. in addition, the separations of the enantiomers of the same analytes on a (r)-n-( 2-naphthyl)-alanine-derived csp was studied. it was indicated that two columns of opposite chirality gave reversed elution orders. |
