英文摘要 | High cell density fermentation of a rAPC producing E. coli strain at 20L scale, purification and antitumor effects of rAPC were studied. One-step and two-step fermentations were employed and the result showed that both expression effi-ciency and biomass of rAPC in two-step fennentation was much higher than those in one-step fermentation. After affinity and gel filtration chromatography, the purity of rAPC reached 98.03% . Inhibitory rate of rAPC on H22 hepatomae could reach 62.2% at the dose of 50 mg/kg per day by i.v application. It was also revealed by experiments that rAPC could inhance the function of B cells by i. p. Result of this paper suggests a promising prospect of large-scale production of rAPC and potential clinical application. |
