英文摘要Due to the particular imaging mode, atomic force microscopy (AFM) provides a new technology for characterizing the topography structures of individual living cells under nearphysiological conditions. However, owing to the lack of adequate immobilization method, using AFM to visualize the topography of mammalian suspension cells is still a big challenge. In this paper, a method for imaging living mammalian suspension cells by AFM was presented. In this method, single B lymphoma suspension cells were mechanically trapped by microfabricated pillars which were coated with positively charged poly-L-lysine. Based on this immobilization, the Rituximab-induced topography changes of individual B lymphoma cells were visualized. The roughness of the cell surface’s local area before and after the stimulation of Rituximab was analysed. The force curves obtained on the cell surface before and after adding Rituximab were also analysed. The experimental results indicate the efficacy of the pillar-based immobilization and will facilitate the research of the physiological activities of living mammalian suspension cells.
